What questions do you ask yourself when developing your plot, characters, etc?

Depends on what it is, and how complicated it is going to get. But outside of that there is the obvious downside to straying into Fantasy/Sci-Fi that it is completely held together by how well you can create the characters and the environments. Anything that takes place in contemporary time is a little more whited on the characters as opposed to the scenery. It is really comes down to how well the reader gets into the story, there are flavors for everyone (Just try not to be vanilla). Being dull and uninteresting are total turnoffs, I don't give a damn how highly rated it is. If it is boring and dull, I won't put much time in it. :D

    Just variations of "why?"

"Why would the character do that?"
"Why is this the course of action they would choose?"
"Why would they feel that way?"
    "Why would that event happen?"

It develops both the plot and the characters at the same time.
